No matter what the situation, no matter how many floors their are, take the stairs, as far as you can, and then take the elevator the rest of the way. Department stores with escalators...climb them.
I work on the 4th floor in a 16 story building. i take the stairs up to 4 every morning...it used to be tough to make it to 4, now i keep going to 6 (our cafeteria) and fill up my water bottle when I get there.

I try to sneak in a few extra calorie burner when I have a chance. For example everytime I have the option of using the escalator/elevator I opt for the stairs. At the parking lot (unless raining) I park furhter away from the entrance to walk a few extra paces.
