I already see several of my favorites mentioned, but haven't seen these yet -

The Goodbye Girl (the one with Richard Dreyfus & Marcia Mason)
The Four Seasons
Young Frankenstein (that's Fronk-en-steen!)
The Thin Man (love the entire series, but the first 2 are the best)
The Full Monty
A Fish Called Wanda
Fierce Creatures
Dave (with Kevin Kline)
Dr. Strangelove (if you like dark comedy)
American Beauty
Monty Python and the Holy Grail
Fargo (more dark comedy)
The Trouble With Harry (really dark comedy)
10 Things I Hate About You (a teen version of The Taming of the Shrew)
Wag the Dog
Airplane!
Mr. Roberts
My Man Godfrey
Dinner at Eight
The Women (the original B&W version, not the crappy attempt at a remake!)

not necessarily funny, but 'feel good' -
Save The Last Dance
The Emperor's Club
The King's Speech
Blossoms in the Desert
Mr. Smith Goes to Washington
Dodsworth

I'm also a big fan of The Marx Brothers movies. Not so much the music, but the comedy is just amazing. Groucho was a genius. Duck Soup and Animal Crackers are probably my favorites.

I usually like Sci-Fi and action comedy, but some of my favorites that were already said: Benny and Joon, Forest Gump, You've Got Mail, O Brother Where Art Thou.
Some I don't think were mentioned: Picture Perfect, Clueless, 13 Going On 30, Ferris Buler's Day Off, Uptown Girls (there's a sad part but overall a good movie).

I loved Mrs Doubtfire too!
I also loved an old movie called "batteries not included". Very cute and feelgood.
My screwy sense of humor fairly reveled in "The Gods Must Be Crazy".
"Princess Bride" fits in there somewhere.
And "Tootsie"!
"Sister Act".
"Rain Man" was pretty good.
