I'm also a big fan of The Marx Brothers movies. Not so much the music, but the comedy is just amazing. Groucho was a genius. Duck Soup and Animal Crackers are probably my favorites.

I usually like Sci-Fi and action comedy, but some of my favorites that were already said: Benny and Joon, Forest Gump, You've Got Mail, O Brother Where Art Thou.
Some I don't think were mentioned: Picture Perfect, Clueless, 13 Going On 30, Ferris Buler's Day Off, Uptown Girls (there's a sad part but overall a good movie).

I know Christmas has come and gone, but lots of the Christmas movies have a strong feel good factor with bits of humour as well. My favourite is probably 'Christmas Choir'.

I loved Mrs Doubtfire too!
I also loved an old movie called "batteries not included". Very cute and feelgood.
My screwy sense of humor fairly reveled in "The Gods Must Be Crazy".
"Princess Bride" fits in there somewhere.
And "Tootsie"!
"Sister Act".
"Rain Man" was pretty good.
